Composition
Active substance:
50.0 g calcipotriol.
Excipients:
alpha-tocopherol 20 g, macrogol stearate 50 mg propylene glycol 100 mg Disodium edetate dihydrate 65 g sodium hydrogen phosphate dihydrate, 260 .mu.g, paraffin liquid (containing about 10 ppm alpha-tocopherol) 50 mg, water 26 mg, paraffin, white soft (contains about 10 ppm alpha-tocopherol) to 1 g
Description:
translucent ointment from white to white with a yellowish tinge.
Product form:
Ointment for external use 0.005%.
30 g of the ointment in an aluminum tube, coated on the inside, with a protective membrane and a plastic screw cap having a tenon on the outer side of the first opening of the protective membrane; 1 tube together with instructions for use in a cardboard box.
Contraindications
Hypersensitivity to Calcipotriol and any excipient the formulation;
Diseases associated with impaired calcium metabolism;
Children up to age 6 years.
Be wary – hypercalcemia, hypercalciuria, vitamin D hypervitaminosis, nephrolithiasis history, age over 65 years, acute renal and hepatic failure.
Indications
Psoriasis vulgaris (including in combination with acitretin, cyclosporine or corticosteroids).
Interaction with other drugs
Dayvoneks® ointment should not be used together with drugs for topical application,
containing salicylic acid
Overdose
Use of the drug in doses exceeding recommended may cause an increase in calcium concentration in blood. In this case, treatment should be discontinued.

Pharmacodynamics:
Calcipotriol is an analog of natural active metabolite of vitamin D3.
Stimulates morphological differentiation and suppresses proliferation of keratinocytes that lies at the heart of its therapeutic effect in psoriasis.
Calcipotriol is an inhibitor of T lymphocyte activation caused by interleukin 1.
The effect on calcium metabolism of calcipotriol is 100 times weaker than with calcitriol.
Pharmacokinetics:
Transdermal absorption of calcipotriol is from 1% to 5% of the dose.
Calcipotriol undergoes rapid biotransformation in the liver to produce pharmacologically active metabolites. The half-life is very short.
Pregnancy and breast-feeding
Safety of the drug during pregnancy and lactation has not been established. It is not known whether calcipotriol in breast milk is released. Should be avoided during pregnancy and lactation

Appointment of patients younger than 18 years can only be short courses and on the skin surface, not exceeding 30% of the body surface.
Should not be applied to large surfaces lesions (more than 30% of the skin surface).
Not recommended for large skin surfaces or severe chronic psoriasis (due to increased absorption intensity and increase the risk of hypercalcemia). If it becomes necessary to use in these forms of psoriasis, it requires careful monitoring of the concentration of calcium in the blood and urine.
Hypercalcemia may occur when exceeding a maximum weekly dose of calcipotriol. To remove the drug in the blood calcium concentration rapidly returning to normal.
