pharmachologic effect
Pharmacological group:
Antifungal agent.
Pharmacodynamics:
Clotrimazole-Teva is an antifungal agent wide spectrum of action for external use. Antifungal effect of clotrimazole active principle (imidazole derivative) is connected with the violation of the synthesis of ergosterol, a component of fungal cell membranes that modifies membrane permeability and cause subsequent lysis of the cells. In low concentrations fungistatic effect, and large – fungicide, not only in proliferating cells. The fungicidal concentrations interacts with mitochondrial and peroxidase enzymes, resulting in increased concentrations of hydrogen peroxide up to a toxic level, which also contributes to the destruction of fungal cells. It is effective against dermatophytes, yeast and molds, as well as the causative agent of multi-colored lichen Pityriasis versicolor (Malazessia furfur) and the pathogen erythrasma. It has antimicrobial action against Gram-positive (staphylococci, streptococci) and Gram-negative bacteria (Bacteroides, Gardnerella vaginalis), as well as against Trichomonas vaginalis. In vitro at a concentration of 0.5-10 ug / ml of clotrimazole inhibits reproduction Corynebacteria family bacteria and gram-positive cocci (except eneterokokkov) and has trihomonatsidnoe action at a concentration of 100 .mu.g / ml.
Pharmacokinetics:
Clotrimazole poorly absorbed through the skin and mucous membranes, and almost no systemic action. The concentration in the deep layers of the epidermis is higher than the minimum inhibitory concentration for dermatophytes. When applied topically clotrimazole concentration in the epidermis is greater than in the dermis and subcutaneous tissue.
