pharmachologic effect
Pharmacological group:
Antiviral agent for topical administration.
Pharmacodynamics:
Antiviral (antiherpetic) agent – a synthetic nucleoside analogue of thymidine.
In infected cells containing viral thymidine kinase, phosphorylation occurs and conversion to atsiklovirmonofosfat. Influenced atsiklovirmonofosfat guanylate cyclase is converted into the diphosphate and under the action of several cellular enzymes – a triphosphate. The high selectivity of action, and low toxicity to humans due to the lack of the enzyme necessary for the formation atsiklovirtrifosfata in intact microorganism cells.
Atsiklovirtrifosfat embedding into synthesized virus deoxyribonucleic acid (DNA), inhibit virus replication. The specificity and the very high selectivity of action is also advantageous due to its accumulation in cells infected with the herpes virus.
Highly active against Herpes simplex virus types 1 and 2; virus that causes chickenpox and zoster (Varicella zoster); Epstein-Barr virus (types of viruses are listed in ascending order of magnitude of the minimum inhibitory concentration (MIC) of acyclovir). Moderately active against cytomegalovirus (CMV).
When herpes prevents new elements disease, reduces the likelihood of cutaneous dissemination and visceral complications accelerates education crusts, reduces pain in the acute phase of herpes zoster.
Pharmacokinetics:
Acyclovir is readily penetrates through the epithelium of the cornea and creates a therapeutic concentration in the aqueous humor. Currently, there are no methods for determining acyclovir in the blood when administered in the form of an eye ointment. Acyclovir at external application is defined only in the urine, and in small quantities. This concentration has no therapeutic value.
